Abstract:

The next generation of antibody immunotherapies demands molecules of unprecedented complexity — from camelid-derived VHH fragments to asymmetric bispecifics — yet conventional discovery and production methods remain bottlenecks. Here, we present our vertically integrated AI-driven drug design (AIDD) platform that generates diverse, high-purity antibody libraries at scale, enabling rapid screening and iterative optimization fueled by real-time experimental feedback. We highlight ongoing platform enhancements, including AI-guided codon optimization and signal peptide selection, which — coupled with our forthcoming TurboCHO 3.0 expression system — deliver multi-fold yield improvements without compromising quality. Two case studies illustrate our approach: first, overcoming the notorious instability and low expression of VHH libraries through framework stabilization and solubility tags; second, deploying analytics to rapidly characterize and suppress deaminated side-products in asymmetric bispecifics — a critical quality attribute often missed until late-stage development. Together, these innovations collapse timelines, reduce attrition, and unlock novel modalities.
